New Oral KRAS G12D Drug: Early Results in Advanced Cancers

Autor:

KRAS G12D is found in some pancreatic, lung and colorectal cancers and has long been difficult to target directly. In a phase 1 trial, 74 previously treated patients with advanced KRAS G12D-mutant solid tumours received the oral drug GFH375. Confirmed tumour shrinkage was seen in about one third of evaluable patients with pancreatic ductal adenocarcinoma or non-small-cell lung cancer. An orthopaedic surgeon following early drug-development studies should distinguish a response signal from comparative evidence: this is an encouraging early signal, but the study had no control group, so it does not yet show how GFH375 compares with standard treatment or whether it improves survival. Serious treatment-related adverse events also occurred, including one treatment-related death from septic shock. The phase 2 part of the study is ongoing.
